Agitation of the solution by rotary or other movement is necessary for dislodging micro- organisms. c) Return the rinse to the bottle. The solution should be examined immediately, if this is not possible, any delay shall be kept to a minimum and the sample shall be cooled quickly to not more than 4oC and maintained between 0oC and 4oC until examined (preferably within 36 hours). 3.2.1.6 Swab Technique 3.2.1.6.1 Accurate Count per Square Inch a) Place cotton-tipped swabs in test-tubes containing 10 ml of distilled water and plug with cotton wool. b) Sterilize in autoclave for 15 minutes at 15 pounds pressure at 120oC. c) Take sterile swabs in tubes to area to be tested. d) Remove sterile swabs from tube and at the same time squeeze excess water from swab against inside of tube. Replace cotton plug on test-tube. e) Usingasterilemetaltemplate,swabanarea of 20 cm2 on the surface to be examined. It is important to keep the swabbing technique uniform e.g. 20 times across from side to side and 20 times up and down, using same amount of pressure. The swab is rotated simultaneously to ensure that all parts of it make contact with the surface under test.
